چکیده

In the last decade, intravital microscopy of breast tumors in mice and rats at single-cell resolution has resulted in important insights into mechanisms of metastatic behavior such as migration, invasion and intravasation of tumor cells, angiogenesis, and immune cells response. We have recently reported a technique to image orthotopic mammary carcinomas over multiple intravital imaging sessions in living mice. For this, we have developed a Mammary Imaging Window (MIW) and optimized imaging parameters for Dendra2 photoswitching and imaging in vivo. Here, we describe the protocol for the manufacturing of MIW, insertion of the MIW on top of a tumor and imaging of the Dendra2- labeled tumor cells using a custom built imaging box. This protocol can be used to image the metastatic behavior of tumor cells in distinct microenvironments in tumors and allows for long term imaging of blood vessels, tumor cells and host cells.

Visualization of Actin Polymerization in Invasive Structures of Macrophages and Carcinoma Cells Using Photoconvertible β-Actin – Dendra2 Fusion Proteins

Actin polymerization controls a range of cellular processes, from intracellular trafficking to cell motility and invasion. Generation and elongation of free barbed ends defines the regions of actively polymerizing actin in cells and, consequently, is of importance in the understanding of the mechanisms through which actin dynamics are regulated. Herein we present a method that does not involve ...

Methodology for Quantitative Characterization of Fluorophore Photoswitching to Predict Superresolution Microscopy Image Quality

Single-molecule localization microscopy (SMLM) image quality and resolution strongly depend on the photoswitching properties of fluorophores used for sample labeling. Development of fluorophores with optimized photoswitching will considerably improve SMLM spatial and spectral resolution. Currently, evaluating fluorophore photoswitching requires protein-conjugation before assessment mandating sp...

Imaging windows for long-term intravital imaging: General overview and technical insights.

Intravital microscopy is increasingly used to visualize and quantitate dynamic biological processes at the (sub)cellular level in live animals. By visualizing tissues through imaging windows, individual cells (e.g., cancer, host, or stem cells) can be tracked and studied over a time-span of days to months. Several imaging windows have been developed to access tissues including the brain, superf...

Energy window setting for optimum Tl-201 cardiac imaging [Persian]

Introduction: Poor sensitivity and poor signal to noise ratio because of low injected thallium dose and presence of scattered photons are the main problems in using thallium in scintigraphic imaging of the heart. Scattered photons are the main cause of degrading the contrast and resolution in SPECT imaging that result in error in quantification. Thallium decay is ve...

An unusual presentation of mammary Paget’s disease with a 12-year duration and no palpable mass

Paget’s disease of the nipple is an unusual manifestation of breast cancer. Most cases of mammary Paget`s disease are associated with an invasive or in situ breast tumor, even with no mass observed in mammography imaging. Very rarely does Paget’s disease occur in the absence of any underlying neoplasia .Herein, we report an 82-year-old woman with an unusual presentation of mammary Paget’s disea...
